NDV-3 is a recombinant protein-based vaccine developed to provide cross-protective immunity against both fungal (Candida species) and bacterial (Staphylococcus aureus) infections. The immunogen in NDV-3 is based on the N-terminal region of the agglutinin-like sequence 3 protein (Als3p) from Candida albicans, formulated with or without alum adjuvant. Als3p is a key adhesin/invasin involved in fungal adhesion, invasion of host tissues, and biofilm formation. The rationale for this design stems from structural homology between C. albicans Als proteins and S. aureus adhesins, enabling cross-reactive immune responses. NDV-3 induces robust humoral (antibody-mediated) and cellular immune responses—particularly Th17-type T-cell responses characterized by IL-17A and IL-22 production—which are critical for protection against both candidiasis and S. aureus infections including methicillin-resistant S. aureus (MRSA). Clinical trials have demonstrated that NDV-3 is safe, well-tolerated, immunogenic in healthy adults, and efficacious in reducing recurrence rates of vulvovaginal candidiasis in women with recurrent disease[1][6][7][8]. A newer formulation known as NDV-3A uses a non-His-tagged version of the antigen[7].
