NDV-F0 is an oncolytic Newcastle Disease Virus (NDV) variant being developed for the treatment of urological cancers, specifically prostate and bladder cancer. It was generated and evaluated by the Oncolytic Viro-Immunotherapy (OVIT) consortium, which includes Leiden University Medical Center, Erasmus Medical Center, and Radboud University Medical Center. NDV-F0 works by preferentially infecting and replicating within tumor cells, leading to direct oncolysis and the induction of systemic anti-tumor immune responses. Preclinical studies have demonstrated its ability to induce immunogenic cell death (ICD) markers such as HSP90, Calreticulin, and HMGB1, and to promote the maturation and activation of monocyte-derived dendritic cells. The virus has shown efficacy in reducing tumor tissue viability in patient-derived human prostate and bladder cancer models, including ex-vivo cultured tumor tissue slices and xenografts.
