nebivolol + valsartan + hydrochlorothiazide is a fixed-dose combination (FDC) therapy that was under clinical investigation for the treatment of essential hypertension. It combines three antihypertensive agents with complementary mechanisms of action: nebivolol, a third-generation, highly selective beta-1 adrenergic receptor antagonist with nitric oxide-mediated vasodilating properties; valsartan, an angiotensin II receptor blocker (ARB) that selectively inhibits the AT1 receptor; and hydrochlorothiazide, a thiazide diuretic that promotes diuresis by inhibiting sodium and chloride reabsorption in the distal convoluted tubule. By targeting the sympathetic nervous system, the renin-angiotensin-aldosterone system (RAAS), and renal fluid balance simultaneously, the combination aims to provide superior blood pressure reduction compared to monotherapy or dual combinations.
