Overview

Neboglamine (formerly known as nebostinel, and also identified by code names CR-2249 and XY-2401) is a small molecule positive allosteric modulator (PAM) that targets the glycine binding site of the N-methyl-D-aspartate (NMDA) receptor. Developed by Rottapharm, the compound is designed to enhance NMDA receptor-mediated neurotransmission, which is hypothesized to be hypoactive in conditions such as schizophrenia. By modulating the glycine site, neboglamine aims to improve cognitive function and alleviate negative symptoms associated with psychiatric disorders. It has also been investigated for its potential in treating cocaine dependence. Preclinical studies in animal models have demonstrated that neboglamine possesses cognition- and memory-enhancing properties. As of the mid-2010s, the drug reached Phase II clinical development for both schizophrenia and cocaine abuse.
