Na-GST-1 is a 24-kDa protein derived from the human hookworm Necator americanus. It functions as a glutathione S-transferase with peroxidase enzymatic activity that catalyzes the conjugation of reduced glutathione to various electrophiles. This hookworm protein belongs to the Nu class of nematode GSTs, characterized by reduced peroxidase activity but elevated binding capacity for heme and related molecules. Na-GST-1 forms homodimers in solution, creating large binding grooves that can bind free heme at high affinity in vitro. In vivo, hookworm GSTs may bind and detoxify heme and hematin byproducts generated during blood degradation. Na-GST-1 is being developed as a vaccine candidate against hookworm infection, with antibodies induced by vaccination postulated to interfere with the digestion of host hemoglobin by adult N. americanus hookworms, thereby impairing their development and survival.
