Overview

Necitumumab is a fully human IgG1 monoclonal antibody that targets the epidermal growth factor receptor (EGFR). It binds with high affinity to the extracellular domain of EGFR, competitively inhibiting ligand binding and preventing receptor dimerization and activation. This blockade disrupts downstream signaling pathways involved in cell proliferation, survival, angiogenesis, invasion, and metastasis. Necitumumab also induces internalization and degradation of EGFR and can mediate antibody-dependent cell-mediated cytotoxicity (ADCC) against tumor cells expressing EGFR. Developed primarily for oncology indications, it is approved for use in combination with gemcitabine and cisplatin as first-line treatment for metastatic squamous non-small cell lung cancer (NSCLC). The drug was developed by Eli Lilly[2][5][7][8].
