Overview

**Necitumumab + gemcitabine + carboplatin** is a combination chemotherapy regimen used in oncology, most notably in the treatment of advanced non-small cell lung cancer (NSCLC). It consists of: - **Necitumumab**: a recombinant human IgG1 monoclonal antibody that targets and inhibits the epidermal growth factor receptor (EGFR), blocking ligand-induced activation and downstream signaling involved in cell proliferation and survival. - **Gemcitabine**: a nucleoside analog (antimetabolite) that is incorporated into DNA during replication, causing chain termination and inhibition of DNA synthesis, resulting in cytotoxicity to rapidly dividing cells like cancer cells. - **Carboplatin**: a platinum-based alkylating agent that forms DNA cross-links, thereby interfering with DNA replication and transcription and ultimately leading to apoptosis of cancer cells. The combination is most frequently used for the first-line treatment of patients with metastatic squamous non-small cell lung cancer who are not candidates for curative surgery or radiation.
