**Necitumumab + mFOLFOX-6** is a combination regimen consisting of **necitumumab**, a recombinant human IgG1 monoclonal antibody targeting the epidermal growth factor receptor (EGFR), combined with **modified FOLFOX-6 (mFOLFOX-6)**. mFOLFOX-6 is a chemotherapy protocol containing oxaliplatin, leucovorin (folinic acid), and fluorouracil (5-FU), widely used for gastrointestinal and colorectal cancers. Necitumumab acts by binding and inhibiting EGFR, interfering with downstream signaling pathways important for tumor cell proliferation and survival. mFOLFOX-6 acts as cytotoxic chemotherapy targeting rapidly dividing cancer cells through DNA crosslinking (oxaliplatin) and inhibition of DNA and RNA synthesis (5-FU, modulated by folinic acid). The combination is under clinical investigation for advanced solid tumors such as metastatic colorectal cancer.
