This regimen is a combination therapy consisting of three agents: **necitumumab**, a recombinant, fully human IgG1 monoclonal antibody targeting the epidermal growth factor receptor (EGFR); **nab-paclitaxel** (albumin-bound paclitaxel), a microtubule inhibitor formulated with albumin nanoparticles; and **carboplatin**, a platinum-based DNA crosslinking agent. This triplet is investigated as first-line therapy for stage IV squamous non-small cell lung cancer (NSCLC). Necitumumab acts as an EGFR antagonist, blocking ligand binding and downstream signaling pathways that promote cell proliferation and survival. Nab-paclitaxel disrupts microtubule dynamics, inhibiting cell division, while carboplatin induces inter- and intra-strand DNA crosslinks, promoting apoptosis in rapidly dividing tumor cells. The combination is not an FDA-approved regimen, but it has progressed to phase II clinical trials and is used in experimental protocols for advanced squamous NSCLC[1][5].
