Nectin-4 CAR-NK cells are an experimental adoptive cell therapy consisting of natural killer (NK) cells genetically engineered to express a chimeric antigen receptor (CAR) targeting Nectin-4 (also known as PVRL4). Nectin-4 is a cell surface adhesion molecule that is highly expressed in several malignancies, including bladder, breast, and lung cancers, while maintaining limited expression in healthy adult tissues, making it an attractive therapeutic target. These CAR-NK cells are designed to combine the innate anti-tumor surveillance of NK cells with the high-precision targeting of a CAR. Preclinical data indicates that these cells can effectively recognize and eliminate Nectin-4-positive bladder cancer cells in both 2D and 3D models, demonstrating enhanced cytotoxicity and tumor infiltration compared to unmodified NK cells.
