Nectin-4-IR700 is a conjugate composed of a monoclonal antibody targeting **Nectin-4**, a tumor-associated antigen highly overexpressed in bladder cancer and other malignancies, covalently linked to **IRDye700DX (IR700)**, a near-infrared photosensitizer. This drug is developed for **near-infrared photoimmunotherapy (NIR-PIT)**, which combines targeted antibody binding and phototoxic cell killing: after systemic administration, Nectin-4-IR700 specifically binds to Nectin-4 on cancer cell surfaces. Subsequent irradiation with near-infrared light activates IR700, causing rapid necrotic cell membrane damage, immunogenic cell death, and tumor destruction. The mechanism is distinct from traditional antibody-drug conjugates, relying on photoactivation rather than cytotoxic payloads[1][2]. In preclinical models, it demonstrates significant antitumor efficacy (suppressed tumor growth, prolonged survival), especially in luminal subtype bladder cancer. Nectin-4-IR700 also enables intraoperative tumor detection via its fluorescence signature, offering both therapeutic and diagnostic utility[1][2].
