Overview

Nectin-4 MMAE ADC refers to a class of antibody-drug conjugates (ADCs) designed to target Nectin-4 (also known as PVRL4), a cell adhesion molecule overexpressed in various solid tumors, including urothelial, breast, lung, pancreatic, and ovarian cancers. These ADCs consist of a monoclonal antibody targeting Nectin-4 conjugated via a cleavable linker to the microtubule-disrupting cytotoxic payload monomethyl auristatin E (MMAE). Upon binding to Nectin-4 on the surface of tumor cells, the ADC is internalized, and the linker is cleaved to release MMAE intracellularly. MMAE then binds to tubulin, inhibiting microtubule polymerization, which leads to G2/M cell cycle arrest and apoptosis. The most prominent and FDA-approved member of this class is enfortumab vedotin (Padcev), developed by Astellas Pharma and Pfizer. Other clinical-stage candidates in this class include Mabwell's 9MW2821 and Corbus Pharmaceuticals' CRB-701.
