NECTIN4×PD-L1 bispecific antibody-drug conjugate is an investigational therapeutic agent that combines the mechanisms of a cytotoxic antibody-drug conjugate (ADC) and an immune checkpoint inhibitor within a single bispecific molecule. Developed by Dong-A ST and AbTis Co., Ltd., the drug utilizes Dong-A ST's bispecific antibody platform with an enhanced knob-into-hole (eKiH) interface to target Nectin-4 and Programmed Death-Ligand 1 (PD-L1). The Nectin-4 arm facilitates the targeted delivery and internalization of cytotoxic payloads—such as monomethyl auristatin E (MMAE) or exatecan—into tumor cells, while the PD-L1 arm blocks the PD-1/PD-L1 axis to restore T-cell activity. This dual-targeting strategy is designed to address tumor heterogeneity and resistance in locally advanced or metastatic urothelial carcinoma (la/mUC) and lung squamous cell carcinoma (LUSC). Preclinical studies have demonstrated potent in vitro cytotoxicity and dose-dependent tumor growth inhibition in xenograft models.
