Overview

Nectin4-7.19 CAR-T is a fourth-generation chimeric antigen receptor (CAR) T-cell therapy designed to target Nectin cell adhesion molecule 4 (Nectin4), a protein overexpressed in various solid tumors including urothelial, lung, and ovarian cancers. Developed by researchers at Wenzhou Medical University and Zhejiang Qixin Biotech, this CAR-T construct is engineered to co-express Interleukin-7 (IL-7) and the chemokine CCL19 (the '7.19' modification). IL-7 is included to enhance T-cell proliferation, survival, and memory formation, while CCL19 promotes the recruitment of endogenous T cells and dendritic cells to the tumor site. This approach aims to overcome the immunosuppressive tumor microenvironment and improve the persistence and infiltration of CAR-T cells in solid tumors. It is currently being evaluated in Phase I clinical trials for Nectin4-positive malignant solid tumors.
