Overview

Nectin4 mCAR-T is a second-generation chimeric antigen receptor (CAR) T-cell therapy developed for preclinical evaluation in immunocompetent mouse models. It features a single-chain variable fragment (scFv) derived from an anti-human Nectin cell adhesion molecule-4 (Nectin4) antibody, fused to murine-derived signaling components including the CD8α hinge and transmembrane regions, the 4-1BB costimulatory domain, and the CD3ζ activation domain. This murine surrogate allows researchers to study the efficacy and safety of Nectin4-targeted CAR-T therapy within a syngeneic immune environment, specifically using C57BL/6 mice and hNectin4-expressing MC38 colorectal cancer cells. Developed by researchers at Wenzhou Medical University and Zhejiang Qixin Biotech, the Nectin4-targeted CAR-T program is currently being evaluated in Phase 1 clinical trials (NCT03932565) for patients with Nectin4-positive malignant solid tumors.
