Overview

Nedaplatin is a second-generation platinum-based antineoplastic (anticancer) drug used in chemotherapy. Structurally, it is a cisplatin derivative in which the two chloride ions of cisplatin are replaced by glycolic acid, resulting in improved water solubility and chemical stability[6][7]. Nedaplatin acts primarily as a DNA-damaging agent; after entering cancer cells, it forms active platinum complexes that bind to purine nucleotides in DNA, causing cross-links and inhibiting DNA replication and transcription. This leads to cell cycle arrest and apoptosis. It also increases oxidative stress by binding to cellular nucleophiles such as glutathione[3]. Nedaplatin can function as a radiosensitizer, making tumor cells more susceptible to radiation therapy[3]. It was initially developed by Nichi-Iko Pharmaceutical Co., Ltd.[1] Its efficacy is similar to that of cisplatin but with reduced gastrointestinal toxicity; however, bone marrow suppression may be more pronounced but remains manageable[4][5][7].
