CDGP + 5-fluorouracil is a combination chemotherapy regimen consisting of nedaplatin (cis-diammine-glycolatoplatinum, CDGP) and 5-fluorouracil (5-FU). Nedaplatin is a second-generation platinum-based small molecule designed to reduce renal and gastrointestinal toxicities compared to cisplatin while maintaining antitumor activity; it works by forming platinum-DNA adducts that inhibit DNA replication and transcription. 5-fluorouracil is a fluoropyrimidine antimetabolite that inhibits thymidylate synthase, thereby disrupting the synthesis of thymidine monophosphate and leading to the inhibition of DNA and RNA synthesis. This combination has been primarily investigated as a concurrent chemoradiotherapy regimen for esophageal squamous cell carcinoma, often utilizing daily low-dose nedaplatin and continuous infusion of 5-FU to enhance the radiosensitizing effects of the treatment.
