Overview

Nefazodone is an antidepressant classified as a phenylpiperazine and serotonin modulator. It is primarily used to treat depression, including major depressive disorder. Its mechanism of action involves inhibiting the neuronal uptake of serotonin and norepinephrine, thereby increasing their levels in the brain. Additionally, nefazodone acts as an antagonist at central 5-hydroxytryptamine type 2 (5-HT2) receptors and alpha1-adrenergic receptors, which may contribute to its therapeutic effects and side effect profile. Unlike selective serotonin reuptake inhibitors (SSRIs), tricyclic antidepressants (TCAs), or monoamine oxidase inhibitors (MAOIs), nefazodone has a distinct chemical structure and pharmacological profile[1][3][4][5]. The drug was originally developed by Bristol-Myers Squibb.
