Overview

Neisseria gonorrhoeae strain FA7527 is an engineered, isogenic mutant of the gonococcus bacterium developed by the National Institute of Allergy and Infectious Diseases (NIAID). This specific strain is a knockout mutant lacking the LptA (lipid A phosphoethanolamine transferase) gene, which is responsible for modifying lipid A with phosphoethanolamine. This modification typically helps the bacteria resist host innate immune defenses, such as cationic antimicrobial peptides. The strain is utilized in Phase 1 experimental human infection models (challenge studies) in healthy male volunteers to investigate the essentiality of LptA for establishing urethral infection and inducing inflammatory responses. The goal of these studies is to validate LptA or related pathways as potential targets for future vaccines or therapeutic interventions against gonorrhea.
