The NEK7 systemic program, developed by Captor Therapeutics, focuses on the creation of small-molecule molecular glue degraders targeting NIMA-related kinase 7 (NEK7) for the treatment of autoimmune and inflammatory diseases. NEK7 is a crucial component of the NLRP3 inflammasome complex; its degradation prevents the activation of the inflammasome and the subsequent release of pro-inflammatory cytokines such as IL-1β. The lead systemic compounds, CPT-513 and CPT-635, function by recruiting an E3 ligase to NEK7 to induce its ubiquitination and proteasomal degradation. These compounds have demonstrated high oral bioavailability (exceeding 95%), picomolar potency, and the ability to maintain sustained target degradation with once-daily dosing in preclinical models. The program is currently being evaluated for indications including inflammatory bowel disease (IBD), gout, and various dermatological diseases.
