Nelipepimut-S is a peptide-based cancer vaccine derived from the extracellular domain of the human epidermal growth factor receptor 2 (HER2/neu or ERBB2) protein. It is a nonapeptide restricted to HLA-A2/A3 and designed to stimulate a cytotoxic T lymphocyte response against tumor cells expressing HER2/neu, which is overexpressed in several cancers including breast cancer. The drug has been primarily investigated for use in preventing recurrence and prolonging survival in patients with breast cancer and other solid tumors that express the HER2 oncoprotein. Nelipepimut-S is typically administered with granulocyte-macrophage colony-stimulating factor (GM-CSF) as an immunoadjuvant to enhance immune response. Clinical trials have shown it can elicit robust anti-HER2 immune responses with minimal side effects; however, development for multiple indications has been discontinued after various phases of clinical trials[1][4][5][6].
