Overview

SD-101 (nelitolimod) is a synthetic class C CpG-oligodeoxynucleotide and investigational immunotherapy drug that acts as a toll-like receptor 9 (TLR9) agonist. It stimulates plasmacytoid dendritic cells (pDCs) to release interferon alpha (IFNα) and mature into efficient antigen-presenting cells, thereby enhancing both innate and adaptive immune responses. The drug is administered intratumorally or via hepatic arterial infusion and has been studied in combination with immune checkpoint inhibitors such as pembrolizumab for the treatment of advanced melanoma, metastatic uveal melanoma with liver metastasis, non-Hodgkin lymphoma, and head and neck squamous cell carcinoma. SD-101 promotes anti-tumor immunity by inducing local production of type I interferons within the tumor microenvironment, leading to cytotoxic T-cell activation against tumor antigens.
