This investigational combination immunotherapy is being developed by TriSalus Life Sciences for the treatment of locally advanced pancreatic ductal adenocarcinoma (PDAC). The regimen consists of nelitolimod (formerly SD-101), a Class C Toll-like receptor 9 (TLR9) agonist, administered in combination with a systemic anti-PD-1 checkpoint inhibitor. A distinguishing feature of this therapy is the use of TriSalus' proprietary Pressure-Enabled Drug Delivery (PEDD) technology via Pancreatic Retrograde Venous Infusion (PRVI) to deliver nelitolimod directly to the tumor site. This regional delivery method is designed to overcome the high interstitial fluid pressure and immunosuppressive tumor microenvironment typical of pancreatic tumors, potentially enhancing the efficacy of the checkpoint blockade by activating the innate immune system and sensitizing the tumor to T-cell mediated attack.
