Overview

This is an investigational combination therapy comprising nelmastobart, trifluridine, tipiracil, and bevacizumab. Nelmastobart is a novel agent under clinical evaluation. Trifluridine and tipiracil are combined in the oral antimetabolite drug Lonsurf; trifluridine is a nucleoside analog that incorporates into DNA causing dysfunction and cell death, while tipiracil inhibits the degradation of trifluridine to maintain its plasma concentration. Bevacizumab is a monoclonal antibody targeting vascular endothelial growth factor (VEGF), inhibiting angiogenesis by preventing VEGF from binding to its receptors on endothelial cells. This combination is being studied for use in patients with metastatic or recurrent colorectal cancer who have progressed after standard therapies such as oxaliplatin and irinotecan[1][3][5][6][8].
