Nemvaleukin alfa is a novel, engineered cytokine and recombinant fusion protein designed to selectively bind to the intermediate-affinity interleukin-2 (IL-2) receptor complex. By doing so, it preferentially activates and expands antitumor CD8+ T cells and natural killer (NK) cells while minimizing the expansion of immunosuppressive regulatory T cells (Tregs). This selective activation leverages the antitumor effects of IL-2 signaling while mitigating toxicities associated with high-affinity IL-2 receptor engagement. Nemvaleukin alfa is being developed primarily for use in cancer immunotherapy, including as monotherapy and in combination with pembrolizumab for advanced solid tumors such as melanoma, ovarian cancer, fallopian tube cancer, peritoneal cancer, squamous cell carcinoma, and other solid tumors. The drug was originally developed by Alkermes and is now under development by Mural Oncology.
