**NEO-212** is an investigational, orally administered small-molecule anticancer agent developed by NeOnc Technologies. It is a single covalent carbamate conjugate of temozolomide and perillyl alcohol, designed to improve central nervous system exposure relative to temozolomide. NEO-212 acts principally as a DNA-alkylating agent, generating cytotoxic DNA damage that can lead to cell-cycle arrest and apoptotic cell death. It has demonstrated preclinical activity in temozolomide-resistant cancer models, including glioblastoma and other gliomas, and is being evaluated in a Phase 1/2 study in progressive IDH-mutant astrocytoma, IDH-wildtype glioblastoma, and uncontrolled brain metastases from selected solid tumors.
