Neo-DCVac is a personalized neoantigen-pulsed autologous dendritic cell vaccine designed for cancer immunotherapy. The therapy involves isolating dendritic cells from the patient, pulsing them with synthetic peptides corresponding to tumor-specific neoantigens identified through genomic analysis of the patient's tumor, and then re-administering these primed cells to stimulate an antigen-specific T-cell immune response against the cancer. Clinical studies have demonstrated that Neo-DCVac is safe, well tolerated, and capable of generating durable antigen-specific lymphocyte responses in patients with advanced cancers such as lung cancer and hepatocellular carcinoma. Early clinical data indicate potential efficacy in controlling disease progression and inducing partial responses or stable disease in heavily pretreated patients[1][2][3][5]. The vaccine is typically administered subcutaneously following pretreatment with cyclophosphamide to enhance immune priming[2][5]. Developers have reported no dose-limiting toxicities or significant adverse events beyond mild injection site reactions and transient rashes[1][2][4].
