NEO-STIM is a proprietary ex-vivo T cell induction and expansion platform developed by Neon Therapeutics (now part of BioNTech). It is designed to identify, prime, and expand patient-specific T cells targeting tumor neoantigens. The process utilizes the RECON bioinformatics engine to predict high-quality neoantigens, which are then used in the NEO-STIM protocol to activate both CD4+ and CD8+ T cell responses from a patient's peripheral blood mononuclear cells (PBMCs). This platform serves as a manufacturing process for personalized adoptive cell therapies, such as NEO-PTC-01 (now BNT221), intended for the treatment of various solid tumors, including melanoma and non-small cell lung cancer (NSCLC).
