NEO212 + FOLFIRI + bevacizumab is a combination cancer therapy consisting of the investigational agent NEO212, the standard chemotherapy regimen FOLFIRI, and bevacizumab. FOLFIRI includes folinic acid (leucovorin), fluorouracil (5-FU), and irinotecan (Camptosar), which work together to halt tumor growth by interfering with DNA synthesis, RNA metabolism, and topoisomerase I activity, leading to cell death of rapidly dividing cells. Bevacizumab is a humanized monoclonal antibody that binds vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis (formation of new blood vessels) critical for tumor growth. Bevacizumab is FDA-approved for several cancers, including metastatic colorectal cancer, when combined with fluorouracil-based chemotherapy[1][2][6]. NEO212 is a conjugate molecule that has demonstrated experimental antitumor activity but is not approved for any indications; its mechanism includes DNA alkylation and possibly other cytotoxic effects.
