NEO212 + ipilimumab is a combination therapy being evaluated for the treatment of primary brain tumors and brain metastases. NEO212 is a small molecule conjugate consisting of the alkylating agent temozolomide (TMZ) covalently linked to perillyl alcohol (POH), a monoterpene designed to enhance the drug's ability to cross the blood-brain barrier and reduce systemic toxicity. Ipilimumab is a fully human monoclonal antibody that targets cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4), a checkpoint molecule that downregulates T-cell activation. By combining the DNA-damaging effects of NEO212 with the immune-stimulating properties of ipilimumab, the regimen aims to overcome the immunosuppressive environment of the central nervous system and improve outcomes for patients with intracranial malignancies.
