Overview

Neoadjuvant chemotherapy + durvalumab + tremelimumab is a combination regimen used in cancer treatment, particularly under investigation for non-small cell lung cancer (NSCLC) and other solid tumors. This regimen combines: - Neoadjuvant chemotherapy (various cytotoxic agents administered before surgery to shrink tumors) - Durvalumab, a monoclonal antibody that targets programmed death-ligand 1 (PD-L1), blocking its interaction with PD-1 and CD80 to enhance T-cell mediated anti-tumor response - Tremelimumab, a monoclonal antibody targeting cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4), another immune checkpoint inhibitor that promotes T-cell activation and proliferation The combination aims to maximize anti-tumor immunity by inhibiting two distinct immune checkpoints while also reducing tumor burden with chemotherapy. Durvalumab and tremelimumab are both developed by AstraZeneca. This approach is being studied in clinical trials for various cancers, including NSCLC[4][7].
