Overview

NeoAg-mBRAF is a synthetic peptide vaccine component representing a shared mutated BRAF-V600E neoantigen. It consists of a 30-amino acid peptide (BRAF 585–614) containing the V600E mutation, with an added N-terminal histidine to facilitate MHC class II restriction. Developed by Craig L. Slingluff, Jr. at the University of Virginia, it is being investigated as part of a multi-component immunotherapy regimen (Mel66 trial) that includes six nonmutated melanoma-specific helper peptides (6MHP), the TLR3 agonist polyICLC, and the CD40-activating antibody CDX-1140. The vaccine aims to stimulate CD4+ T cell immune responses against the BRAF V600E neoantigen in patients with high-risk cutaneous or uveal melanoma in the adjuvant setting.
