Overview

NeoAg-VAX is a personalized cancer vaccine designed to stimulate the immune system to recognize and attack tumor-specific neoantigens in patients with colorectal cancer, particularly those with microsatellite stable (MSS) metastatic disease. The vaccine is tailored for each patient by identifying unique neoantigens present in their tumor cells, which are then formulated into a vaccine to induce robust antigen-specific T cell responses. Clinical trials have demonstrated that NeoAg-VAX has a favorable safety profile, induces strong immunogenicity, and leads to dynamic changes in the tumor microenvironment that may help overcome immune evasion. It is being evaluated both as monotherapy and in combination with pembrolizumab (Keytruda), an anti-PD-1 checkpoint inhibitor[1][4][8].
