A **neoantigen peptide-pulsed dendritic cell vaccine** is a form of personalized immunotherapy where autologous dendritic cells are isolated from the patient, matured and loaded ex vivo with synthetic peptides corresponding to tumor-specific neoantigens identified by sequencing the patient's tumor. The vaccine is then re-administered to the patient to prime the immune system, especially cytotoxic T cells, to recognize and attack cancer cells harboring those mutations. This approach is under clinical investigation in several solid tumors, including glioblastoma, lung cancer, hepatocellular carcinoma, and pancreatic cancer. Neoantigen DC vaccines have demonstrated feasibility, safety (primarily grade 1–2 adverse events), immunogenicity (eliciting neoantigen-specific T cell responses), and promising early efficacy in pilot human studies. The vaccines are highly individualized, with peptide antigens tailored to each patient's tumor mutation profile[1][3][5][6][7].
