Overview

The neoantigen-pulsed autologous dendritic cell vaccine is a personalized cancer immunotherapy developed by Peking University Cancer Hospital. This therapeutic approach involves identifying patient-specific tumor mutations (neoantigens) through whole-exome and RNA sequencing of tumor and normal tissues. Synthetic peptides or mRNA corresponding to these unique neoantigens are then used to 'pulse' or load autologous dendritic cells (DCs) derived from the patient's own peripheral blood mononuclear cells. These mature, antigen-loaded DCs are subsequently re-infused into the patient, where they act as professional antigen-presenting cells to prime and activate neoantigen-specific CD4+ and CD8+ T cells. This targeted immune response is designed to recognize and eliminate tumor cells while minimizing damage to healthy tissue. The vaccine is currently being evaluated in clinical trials for various advanced solid tumors, including gastric, pancreatic, and biliary tract cancers.
