Overview

This is a personalized cancer vaccine consisting of autologous (patient-derived) dendritic cells pulsed with synthetic peptides representing patient-specific tumor neoantigens. Developed by the Universidad Nacional de Colombia, the therapy is specifically designed for patients with triple-negative breast cancer (TNBC). The manufacturing process involves genomic sequencing of the patient's tumor to identify unique non-synonymous mutations, followed by the synthesis of 25-amino acid peptides incorporating these mutations. These peptides are then loaded onto the patient's own dendritic cells *ex vivo*. When administered intradermally, these 'pulsed' dendritic cells present the neoantigens to the patient's immune system, priming and activating CD4+ and CD8+ T cells to recognize and destroy tumor cells expressing the specific mutations. The therapy is currently being evaluated in a Phase I clinical trial (NCT06195618) to assess its safety and immunogenicity in the neoadjuvant setting.
