Overview

Neoantigen-targeted dendritic cell T-cell therapy is a personalized cellular immunotherapy developed by the Department of Biotherapy and Cancer Center at West China Hospital, Sichuan University. This therapeutic approach involves a dual-component strategy: a dendritic cell (DC) vaccine and adoptive T-cell therapy. First, patient-specific tumor mutations (neoantigens) are identified through next-generation sequencing (NGS) and bioinformatic analysis of tumor biopsies. Autologous dendritic cells are then loaded (pulsed) with these neoantigens to create a vaccine designed to prime and activate the patient's immune system. Simultaneously, neoantigen-reactive T cells (NRTs) are identified, isolated, and expanded ex vivo. The combined administration of the DC vaccine and the expanded T cells aims to overcome the immunosuppressive tumor microenvironment and induce a potent, highly specific cytotoxic response against tumor cells expressing those unique mutations. This therapy is primarily being investigated for advanced solid tumors, including melanoma, bladder cancer, and colorectal cancer.
