The neoantigen-targeted ppDC vaccine is an investigational personalized immunotherapy developed by researchers at the UCLA Health Jonsson Comprehensive Cancer Center. It is specifically designed to treat H3 G34-mutant diffuse hemispheric glioma, a rare and aggressive brain tumor typically found in adolescents and young adults. The therapy involves harvesting a patient's own dendritic cells and pulsing them with peptides (ppDC) that represent neoantigens—unique proteins resulting from mRNA splicing changes caused by the histone H3 G34R mutation. When administered intradermally alongside the adjuvant Poly ICLC (a TLR3 agonist), the vaccine aims to prime and activate T lymphocytes to recognize and destroy tumor cells expressing these specific neoantigens.
