Overview

A combination immunotherapy consisting of a personalized neoantigen vaccine and the adjuvant poly-ICLC. The neoantigen vaccine is designed using patient-specific tumor mutations to generate peptides (neoepitopes) that are unique to the individual's cancer. These peptides are administered with poly-ICLC (Hiltonol), a synthetic double-stranded RNA that acts as an immune adjuvant by mimicking viral infection and activating innate immune responses through pattern recognition receptors on antigen-presenting cells. This combination aims to stimulate robust CD8+ and CD4+ T cell responses against tumor cells presenting these neoantigens, enhancing anti-tumor immunity. Poly-ICLC boosts dendritic cell activation and antigen presentation, further amplifying the adaptive immune response. This approach is under investigation for various cancers including melanoma, kidney cancer, glioma, and others[1][2][3][4][5][6][8].
