This combination therapy is an experimental immunotherapy regimen being evaluated for the treatment of advanced or unresectable sarcomas, including osteosarcoma and soft tissue sarcoma. It consists of three distinct components: **SarcVac**, a personalized tumor neoantigen vaccine; **tumor-specific lymphocytes (TSLs)**, which are autologous immune cells expanded ex vivo and reinfused into the patient; and an **anti-PD1 antibody**, a checkpoint inhibitor. The vaccine is designed to prime the patient's immune system to recognize unique mutations (neoantigens) present on their specific tumor cells. The reinfused lymphocytes provide a robust population of effector cells to directly attack the tumor, while the PD-1 antibody prevents the tumor from evading the immune response by blocking the PD-1/PD-L1 inhibitory pathway. This triple combination aims to overcome the immunosuppressive microenvironment of advanced sarcomas and enhance anti-tumor efficacy in patients who have failed standard treatments.
