Neobe Therapeutics is developing a platform of engineered live biotherapeutic products (LBPs) using non-pathogenic, commensal-derived bacteria. These bacteria are programmed with synthetic biosensors to colonize the tumor microenvironment (TME) by responding to specific environmental cues such as hypoxia and low pH. Once localized within the tumor, the bacteria are engineered to locally secrete enzymes that degrade the extracellular matrix (ECM), specifically targeting components like hyaluronan via the secretion of hyaluronidase. This remodeling of the TME is intended to break down dense fibrotic barriers, thereby improving the infiltration of immune cells and the delivery of co-administered therapies such as checkpoint inhibitors, CAR-T cells, and antibody-drug conjugates. The platform is currently in preclinical development for the treatment of solid tumors, including breast, pancreatic, and colorectal cancers, with the goal of priming tumors that are otherwise resistant to immunotherapy.
