NeoBOMB1 is a novel DOTA-coupled peptide antagonist of the gastrin-releasing peptide receptor (GRPR), designed for theranostic use in oncology. It can be radiolabeled with isotopes such as gallium-68 (for PET imaging) and lutetium-177 (for targeted radionuclide therapy). NeoBOMB1 binds with high affinity to GRPR, which is overexpressed in several tumor types including breast cancer, prostate cancer, and gastrointestinal stromal tumors. Unlike earlier bombesin analogs that acted as agonists and caused unwanted side effects, NeoBOMB1 functions as an antagonist, offering improved metabolic stability and reduced side effects. Its mechanism involves targeting GRPR-expressing cells to deliver diagnostic or therapeutic radioisotopes directly to tumors. The drug has demonstrated excellent tumor targeting, favorable pharmacokinetics, high metabolic stability in vivo, and promising safety profiles in preclinical models and early clinical trials[7][4][3][8]. Developers include Erasmus University Rotterdam and the National Center for Scientific Research Demokritos; Advanced Accelerator Applications is a key developer for clinical translation[3].
