Overview

Neohesperidin dihydrochalcone (NHDC) is a semi-synthetic flavonoid sweetener derived from citrus fruits, primarily known for its intense sweetness and use as a food additive (E959). Beyond its application in the food industry, NHDC has emerged as a potential therapeutic agent due to its antioxidant and anti-inflammatory properties. Recent research, such as that presented at AACR 2026, identifies NHDC as a potent inhibitor of ferroptosis, a form of iron-dependent regulated cell death. It acts by targeting the System Xc− pathway, mitigating the accumulation of lipid reactive oxygen species (ROS) and modulating the expression of ferroptosis-related proteins like SLC7A11 and SLC3A2. This mechanism suggests potential applications in treating various ferroptosis-related pathologies, including cancer and neurodegenerative diseases.
