NeoPep is a personalized neoantigen-specific peptide vaccine system developed by Hangzhou NeoVax Biotechnology for the treatment of newly diagnosed glioblastoma and glioma. The vaccine regimen consists of two distinct components: NPVAC1, a "warehouse" vaccine containing five peptides derived from Human Cytomegalovirus (HCMV) antigens, and NPVAC2, a personalized vaccine comprising 20 de novo synthesized peptides tailored to the individual patient's tumor mutanome and HLA ligandome. Administered subcutaneously alongside the TLR3 agonist Poly-ICLC (Hiltonol) and standard temozolomide chemotherapy, the vaccine is designed to stimulate a robust, multi-epitope T-cell response to overcome the low mutational burden and immunosuppressive environment characteristic of glioblastoma.
