Overview

NeoSTX + bupivacaine + epinephrine is an investigational combination of three agents designed for prolonged local anesthesia. Neosaxitoxin (NeoSTX) is a potent site-1 sodium channel blocker derived from marine toxins, which produces long-lasting sensory and motor nerve block by inhibiting action potential propagation in nerves. Bupivacaine is a widely used amino amide local anesthetic that blocks voltage-gated sodium channels to prevent nerve impulse transmission. Epinephrine acts as a vasoconstrictor, reducing systemic absorption of the anesthetics and prolonging their local effect while decreasing toxicity risk. The combination has been shown in preclinical and early clinical studies to provide significantly longer duration of anesthesia compared to standard agents alone or dual combinations, with improved safety profile due to reduced systemic exposure and side effects[1][2][4][5]. This formulation is being developed primarily for use in surgical procedures requiring extended regional anesthesia.
