Overview

neoTCR-T cells (specifically NeoTCR-P1) is a personalized, autologous, neoantigen-specific T-cell receptor (TCR) engineered T-cell therapy developed by PACT Pharma. The therapy is designed to target patient-specific tumor neoantigens, which are unique mutations presented by human leukocyte antigens (HLAs) on the surface of tumor cells. Using a non-viral, CRISPR-Cas9-mediated precision genome-editing process, the patient's endogenous T-cell receptors are knocked out, and cloned neoepitope-specific TCRs isolated from the patient's peripheral blood are knocked in. This precision engineering creates a customized population of CD8+ and CD4+ T cells capable of trafficking to and destroying the patient's specific tumor cells. NeoTCR-P1 was evaluated in a Phase 1a/1b clinical trial for various advanced or metastatic solid tumors, including colorectal cancer, breast cancer, and melanoma, but development was suspended following a strategic restructuring by PACT Pharma.
