Overview

NER1006 (brand name Plenvu) is a low-volume, 1-liter polyethylene glycol (PEG)-based osmotic laxative developed by Norgine and commercialized in the United States by Salix Pharmaceuticals. It is specifically indicated for cleansing of the colon in preparation for colonoscopy in adults. NER1006 is the first FDA-approved 1-liter PEG-based bowel preparation, designed to improve patient compliance by reducing the total volume of fluid required compared to traditional 2-liter or 4-liter PEG solutions. The formulation is provided as two doses: Dose 1 contains PEG 3350, sodium sulfate, sodium chloride, and potassium chloride; Dose 2 contains PEG 3350, sodium chloride, potassium chloride, and a high dose of ascorbate (ascorbic acid and sodium ascorbate). Its mechanism of action is osmotic, where the PEG and ascorbate components create an osmotic gradient that draws water into the intestinal lumen, increasing stool volume and stimulating peristalsis to facilitate thorough bowel cleansing.
