Overview

Neramexane is a small molecule drug structurally related to memantine. It acts as an uncompetitive antagonist of the N-methyl-D-aspartate (NMDA) receptor channel and also antagonizes the alpha9-alpha10 nicotinic acetylcholine receptor. Neramexane was developed primarily for central nervous system disorders including tinnitus and Alzheimer's disease. Its mechanism involves neuroprotection by inhibiting excessive glutamate-mediated excitatory neurotransmission and modulating cholinergic signaling. Clinical trials have investigated its use in tinnitus (up to phase III), Alzheimer's disease, pain (acute or chronic), drug addiction/alcohol dependence, and other CNS conditions[1][2][4][5][7].
