neratinib + fulvestrant + eribulin is a combination therapy being investigated in a Phase 2 clinical trial sponsored by Xijing Hospital for the treatment of hormone receptor-positive (HR+) and human epidermal growth factor receptor 2-positive (HER2+) advanced breast cancer, particularly in patients who have progressed on trastuzumab deruxtecan (T-DXd). The regimen combines three distinct mechanisms: neratinib, an irreversible tyrosine kinase inhibitor of HER1 (EGFR), HER2, and HER4; fulvestrant, a selective estrogen receptor degrader (SERD) that competitively binds to estrogen receptors and promotes their degradation; and eribulin, a non-taxane microtubule dynamics inhibitor that induces cell cycle arrest by inhibiting microtubule growth. This triplet approach is designed to provide a comprehensive blockade of the signaling pathways driving tumor growth and resistance in triple-positive breast cancer.
