Overview

Nerve growth factor (NGF) is a neurotrophin protein that plays a critical role in the growth, maintenance, and survival of sympathetic and sensory neurons. In adults, NGF regulates neuronal regeneration after injury and modulates pain perception by binding primarily to the tropomyosin receptor kinase A (TrkA) with high affinity and to p75 with low affinity. Activation of these receptors initiates intracellular signaling pathways involved in cell survival, proliferation, or apoptosis. NGF is elevated in various chronic pain conditions such as osteoarthritis, interstitial cystitis, arthritis, pancreatitis, cancer pain, and diabetic neuropathy. Recombinant or exogenous NGF has been investigated for its potential therapeutic effects in neurological disorders including Alzheimer's disease and traumatic brain injury[2][3][4][6]. However, most clinical development has focused on inhibitors of NGF for chronic pain management.
